3D structural modeling of bacterial proteins using metagenome sequences and coevolution-based contact prediction

3D protein structures provide detailed insight into biochemical pathways and metabolic functions that will ultimately lead to a predictive understanding of complex biological systems behavior. However, experimental structure determination is often challenging, particularly for membrane proteins. Thus, new strategies must be developed, and computational approaches will play a vital role. Recently, three techniques—metagenomics, coevolution analysis, and de novo protein structure prediction—were combined to produce accurate structures of a large number of proteins that were previously intractable to model (S. Ovchinnikov et al., Science, 2017). I will describe the application of this approach in developing a structural model of a bacterial membrane-bound protein complex.
